چکیده

A salt-extractable low-molecular-weight fraction has been held responsible for the resistance of cartilage to invasion by malig nant cells. To test this hypothesis, we have confronted in vitro fragments from bovine articular cartilage, from bovine nasal septum, from chick embryonic tibia, or from human knee menis cus with cells from the following malignant lines: LICR-HN-4 human squamous cell carcinoma; B16BL6 mouse melanoma; Hu-456 human transitional cell carcinoma; TE-85 and SAOS-2 human osteosarcoma; and MO4 mouse fibrosarcoma. Human embryo lung cells and chick embryonic heart cells were used as nonmalignant counterparts. Confronting cultures using living car tilage and cartilage extracted with 3 M guanidinium hydrochloride were examined light microscopically and ultrastructurally after 1 to 14 days. Neither invasion of malignant cells into the matrix of living or salt-extracted cartilage nor breakdown of collagen was observed in these cultures. In contrast, both malignant and nonmalignant cells occupied preexisting spaces in the matrix, namely, cut chondrocyte lacunae, cartilage canals, and fibrillation clefts. We concluded from these experiments that salt extraction did not alter the resistance of cartilage to invasion by malignant cells in vitro. This conclusion does not support the opinion that salt-extractable factors are solely responsible for the resistance

Designing of Human Cartilage Tissue, by Differentiation of Adipose-Derived Stem Cells With BMP-6 in Alginate Scaffold

Purpose: In the present study the effect of BMP-6 was investigated on chondrogenesis of adiposederived stem cells. Materials and Methods: Mesenchymal stem cells derived from subcutaneous adipose tissue were cultured on alginate scaffold to induce chondrogenesis in experimental group, with chondrogenic medium having BMP-6 growth factor for 3 weeks. In control group medium without BMP-6 was appli...

Research Article: Do derived whitecheek shark proteins motivate T cells to fight cancer cells? A case study in using SW742 cell line

Shark cartilage is considered as a natural dietary supplement consisting of anti-angiogenic, immunostimulatory, and anti-inflammatory characteristics. Therefore, this study was designed to inspect the possibility that whitecheek shark (Carcharhinus dussumieri) cartilage proteins motivate expression of NKG2D, CXCR3, NKP46, and NKP44 receptors on natural killer cells and their activities against ...

Evaluation of the Effect of Ursolic Acid Compared with Ibuprofen and Dexamethasone in Synoviocyte Model for Osteoarthritis: an Ex Vivo Study

Background: Osteoarthritis is one of the most commonly diagnosed malignant diseases in the world and was previously recognized as a degenerative joint disease characterized by cartilage deformity and the formation of new bones at the edges of the joint. But this disease is not just a degenerative disease but also a biomechanical, biochemical, and cellular phenomenon. Common treatments include s...

Small Molecule-BIO Accelerates and Enhances Marrow-Derived Mesenchymal Stem Cell in Vitro Chondrogenesis

Background: Hyaline cartilage defects exhibit a major challenge in the field of orthopedic surgery owing to its limited repair capacity. On the other hand, mesenchymal stem cells (MSCs) are regarded as potent cells with a property of cartilage regeneration. We aimed to optimize marrow-derived MSC chondrogenic culture using a small bioactive molecule referred to as BIO. Methods: MSCs from the ma...
